Description

Trans-cyclooctene-lysine is a synthetic amino acid derivative that integrates the bioorthogonal reactivity of trans-cyclooctene (TCO) with the side chain of lysine, a naturally occurring amino acid. TCO-lysine can be genetically encoded into proteins using engineered tRNA synthetases, allowing for the site-specific incorporation of TCO groups into proteins at specific lysine residues. Once incorporated, the TCO group can undergo a highly selective reaction with tetrazine-functionalized probes, enabling site-specific protein labeling in live cells. This method is widely used for labeling proteins with fluorophores, biotin, or other tags for imaging or purification purposes. Because the TCO-tetrazine reaction is bioorthogonal, it can occur in complex biological environments without interfering with natural biological processes, making it ideal for live-cell studies.
