TCO-modified ncAAs (Trans-Cyclooctene-modified non-canonical amino acids) are specialized amino acids that have been functionalized with trans-cyclooctene (TCO) groups. ncAAs are amino acids that are not commonly found in the standard genetic code. They can include variations in side chains, backbone structures, or functional groups that enhance the properties of proteins, such as increased stability, novel functionalities, or enhanced binding capabilities. TCO-modified ncAAs can be used to deliver therapeutic agents specifically to target proteins or cells. By attaching drugs to tetrazine-labeled carriers, these carriers can be selectively directed to TCO-modified proteins, enhancing the specificity and efficacy of treatments. This targeted approach can minimize off-target effects and improve the therapeutic index of drugs.
