Description

Mal-PEG4-Val-Ala-PAB is a cleavable ADC linker consisting of a maleimide group (Mal), a valine-alanine dipeptide (Val-Ala), and a p-aminobenzyloxycarbonyl (PAB) protecting group. The maleimide group is highly reactive with sulfhydryl groups on proteins and peptides, allowing it to form stable thioether linkages between the linker and the target molecule. The Val-Ala dipeptide provides a spacer arm that can help to minimize steric hindrance and improve the accessibility of the maleimide group to the target molecule. The PAB protecting group can be removed under mild conditions to reveal an amino group, which can be used for further conjugation or drug loading. PEG space improve compound’s aqueous solubility.
