Description

Biotin-HPDP or (3aS,4S,6aR)-hexahydro-2-oxo-N-[6-[[1-oxo-3-(2-pyridinyldithio)propyl]amino]hexyl]-1H-thieno[3,4-d]imidazole-4-pentanamide is widely used for thiol-specific labeling of proteins and peptides. The HPDP group reacts selectively with free thiols (-SH) in proteins under mild conditions, forming stable disulfide bonds. This allows for site-specific attachment of biotin to cysteine residues on proteins, enabling subsequent purification, detection, or immobilization using streptavidin or avidin. In addition, the reversible nature of the disulfide bond formed by HPDP allows for the development of redox-sensitive probes and sensors. Biotin-HPDP conjugates can be used to study thiol redox status in biological systems, such as redox signaling pathways, oxidative stress responses, or protein folding dynamics.
