Biotin-VAD-FMK or methyl (3S,6S,9S)-3-(2-fluoroacetyl)-9-isopropyl-6-methyl-5,8,11,18-tetraoxo-22-((3aS,4S,6aR)-2-oxohexahydro-1H-thieno[3,4-d]imidazol-4-yl)-4,7,10,17-tetraazadocosanoate consists of biotin conjugated to the VAD peptide sequence, which in turn is linked to the FMK group. Biotin, also known as Vitamin B7, is a small molecule that binds strongly to avidin and streptavidin proteins VAD is a peptide sequence that mimics the substrate specificity of caspases, a family of protease enzymes crucial in apoptosis pathways. FMK is a reactive group used to irreversibly inhibit the catalytic activity of caspases. FMK derivatives are commonly used as caspase inhibitors to study apoptosis and related processes. Biotin-VAD-FMK is used to study apoptosis by labeling and inhibiting active caspases in cells undergoing programmed cell death. It allows researchers to identify and quantify apoptotic cells and to investigate the role of caspases in apoptosis signaling pathways.
