Lificiguat (also known as YC-1) is a small molecule with potent vasodilatory and anti-fibrotic properties. It is a derivative of the natural compound 3-(5′-hydroxymethyl-2′-furyl)-1-benzylindazole (YC-1), originally identified for its ability to activate soluble guanylate cyclase (sGC), a key enzyme involved in the nitric oxide (NO) signaling pathway. YC-1 has been investigated for its potential therapeutic effects in various conditions, particularly those involving endothelial dysfunction and fibrosis. Lificiguat’s vasodilatory effects make it a promising candidate for the treatment of pulmonary arterial hypertension (PAH), a condition characterized by elevated blood pressure in the arteries of the lungs, leading to right heart failure.
