Strained-trans-cyclooctene (s-TCO) is a highly reactive form of trans-cyclooctene (TCO), which has been further structurally modified to increase the ring strain, thereby enhancing its reactivity in bioorthogonal chemistry, particularly in the inverse electron demand Diels-Alder (IEDDA) reaction with tetrazines. s-TCO reacts much faster with tetrazines compared to conventional TCO due to the higher strain in the ring. This makes it especially useful in time-sensitive applications, such as real-time imaging or fast biochemical labeling.
