T4L-TCO is a protein-reactive compound that combines the reactive properties of trans-cyclooctene (TCO) with the specificity of T4 lysozyme (T4L). T4 lysozyme is an enzyme derived from the T4 bacteriophage, known for its ability to lyse bacterial cell walls. In T4L-TCO, T4L has been engineered to carry a reactive TCO group. The TCO group in T4L-TCO is a bicyclic compound that is highly reactive in bioorthogonal click chemistry, particularly with tetrazines in the inverse electron demand Diels-Alder reaction. The combination of T4L and TCO provides a protein-reactive version of the cyclooctene that can specifically interact with targeted biomolecules.
