Trans-cyclooctene-protected SIINFEKL (TCO-SIINFEKL) refers to the peptide SIINFEKL that has been modified with a trans-cyclooctene (TCO) group. SIINFEKL is an 8-amino acid peptide (Ser-Ile-Ile-Asn-Phe-Glu-Lys-Leu) that binds specifically to the MHC class I molecule H-2Kb, making it a model antigen for studying CD8+ T-cell responses. TCO-SIINFEKL can be labeled in live cells or organisms with tetrazine-containing fluorescent dyes or imaging agents, allowing researchers to track antigen presentation, T-cell recognition, and immune responses in real time. In addition, the TCO modification enables the tracking of T-cell activation in response to the SIINFEKL epitope, helping to understand the dynamics of immune responses, particularly in cancer immunology and vaccine development.
